Heavy-chain receptor editing unbound.

چکیده
With all of the certainty of death and taxes, questions regarding the mechanisms of immunological tolerance remain fixtures across the landscape of immunology. From Paul Ehlirch’s “horror autotoxicus” (reviewed in ref. 1) to the current synthesis of tolerance by apoptosis, anergy, and receptor editing (2), the mechanisms of immunological tolerance remain, despite substantial research effort, enigmatic. In PNAS, Kumar et al. (3) and Sun et al. (4) refine our understanding of receptor editing, a principle component of immune tolerance, by demonstrating that at least one form of this tolerance pathway is not driven by reaction with self-antigens.

Molecular mechanisms involved in receptor editing at the Ig heavy chain locus.
In receptor editing, a phenomenon that has recently come to light and into favor, a rearranged VDJ or VJ gene segment encoding a variable region of an Ig chain is replaced by another. In this commentary, the molecular mechanisms involved in the editing process are examined in some detail. Editing is most likely mediated by the same V(D)J recombinase activity responsible for the formation of the...
متن کاملEditing Anti-DNA B Cells by Vλx
Receptor editing is performed by replacement of Vkappa genes that contribute to autoreactivity. In addition, the Ckappa locus can be deleted by Vkappa rearrangement to intronic or 3' of Ckappa RS sequences (also referred to as kappa deletion elements). B cells that delete the Ckappa can then express lambda light chains. However, the lambda locus, either of man or mouse, does not allow V gene re...
متن کاملPositive Selection of Natural Poly-Reactive B Cells in the Periphery Occurs Independent of Heavy Chain Allelic Inclusion
Natural autoreactive B cells are important mediators of autoimmune diseases. Receptor editing is known to play an important role in both central and peripheral B cell tolerance. However, the role of allelic inclusion in the development of natural autoreactive B cells is not clear. Previously, we generated μ chain (TgV(H)3B4I) and μ/κ chains (TgV(H/L)3B4) transgenic mice using transgene derived ...
متن کاملB cell deletion, anergy, and receptor editing in "knock in" mice targeted with a germline-encoded or somatically mutated anti-DNA heavy chain.
To study the relative contributions of clonal deletion, clonal anergy, and receptor editing to tolerance induction in autoreactive B cells and their dependence on B cell receptor affinity, we have constructed "knock in" mice in which germline encoded or somatically mutated, rearranged anti-DNA heavy (H) chains were targeted to the H chain locus of the mouse. The targeted H chains were expressed...
متن کاملLight chain editing in kappa-deficient animals: a potential mechanism of B cell tolerance
The genetic organization of the kappa and lambda light chain loci permits multiple, successive rearrangement attempts at each allele. Multiple rearrangements allow autoreactive B cells to escape clonal deletion by editing their surface receptors. Editing may also facilitate efficient B cell production by salvaging cells with nonproductive light chain (L chain) rearrangements. To study receptor ...
